The Study of Human CD4+ T-cells Using Class II Tetramers

چکیده

Esta revisión habla sobre la emergencia de los tetrámeros como reactivos para el estudio de la inmunidad humana. Los tetrámeros del complejo mayor de histocompatibilidad (MHC) han generado nuevas e interesantes posibilidades para la caracterización de las células T específicas de antígeno. Estos tetrámeros tienen la capacidad única de marcar y subsecuentemente facilitar el aislamiento o la estimulación de células T con una sola restricción de péptido-MHC. Por eso los tetrámeros tienen el potencial de refinar o incluso redefinir las respuestas inmunitarias T tal y como las entendemos actualmente. Además de una revisión breve sobre la historia y el desarrollo de los tetrámeros, este artículo ofrece un resumen de la producción de tetrámeros de clase II usando un sistema de expresión en Drosophila, y apunta varios métodos para su aplicación. Las similitudes y diferencias más importantes entre los tetrámeros de clase II, técnicas tradicionales y otras técnicas alternativas también serán detalladas, junto con una previsión de las direcciones futuras de esta tecnología.
